SUMMARY
The eyes begin to develop as a pair of puffs on either side of the forebrain that will develop into the optic vesicles at the end of the fourth week of development. The optic vesicles come into contact with the superficial ectoderm and induce the formation of the lens. When the optic vesicle begins to invaginate to raise the pigmented and neural layers of the retina, the lens plaque invaginates to form the lens vesicle. Through a groove located in the inferior aspect of the optic vesicle, the choroidal fissure, the hyaloid artery (later the central artery of the retina) enters the eye. The nerve fibers of the eye also occupy this groove on its way to the visual areas of the brain. The cornea is formed from (1) a layer of superficial ectoderm; (2) the stroma, which maintains continuity with the sclera, and (3) an epithelial layer, which limits the anterior chamber.
PAX6, a master gene for eye development, is expressed in the common optical field at the neural plate stage. The optical field is divided into two optical primordia by the action of SHH, which generates positive regulation of PAX2 expression in the optical stems, while regulating PAX6 negatively, limiting gene expression to the optic cup and the lens. The epithelium-mesenchyme interactions between the ectoderm of the future lens, the optic vesicle and the surrounding mesenchyme then regulate the differentiation of the lens and the optic cup.
