Its a method which is used for research purposes to determine the minimum inhibitory concentration of an antimicrobial agent which is usually an antibody. What do we do in this method.... We prepare agar plates.... And The agar surface is inoculated by spreading a particular volume of the microbial inoculum, over the entire agar surface. The wells are then made in the gel with a well punture... Into these wells, a known volume and concentration of antibiotic is added..... The antibiotic diffuses from the well.... And the diffusion of antibiotic leads to inhibition of bacterial growth, in the vicinity of the puntured well And the area upto which the antibiotic shows its effect... Is reffered as zone of inhibition....
