Materials required
Spinach/Geranium Leave sample (250mg)
Acetone (80%)
Measuring cylinder
weigh balance
Funnel
Filter paper
Pestle and mortar
Gloves
Mask
Procedure
Method1
Step1. Grind the leaves with acetone via pestle and mortar
Step2. Allow the preparation to settle in the dark
Step3. Filter by using filter paper
Step4. Let acetone evaporate

Extraction of chlorophyll (Arnon, 1949) One gram of finely cut fresh leaves were taken and ground with 20 – 40ml of 80% acetone. It was then centrifuged at 5000 –10000rpm for 5mins. The supernatant was transferred and the procedure was repeated till the residue becomes colorless. The absorbance of the solution was red at 645nm and 663nm against the solvent (acetone) blank.
