IN THIS LECTURE I DISCUSS about cAMP Pathway G-Protein Coupled Receptor (GPCR) .
In this video we have discussed about the Adenyl Cyclase cAMP Pathway which is one transducer mechanism of GPCR
In the field of molecular pharmacology, the cAMP-dependent pathway, also known as the adenylyl cyclase pathway, is a G protein-coupled receptor-triggered signaling cascade used in cell communication.
cAMP was the first second messenger to be identified. Its three main effectors are PKA (which phosphorylates numerous metabolic enzymes), EPAC (a guanine-nucleotide-exchange factor), and cyclic-nucleotide-gated ion channels.
Most ACs (soluble bicarbonate-regulated ACs are the exception) are activated downstream from G-protein-coupled receptors (GPCRs) such as the β adrenoceptor by interactions with the α subunit of the Gs protein (αs). αs is released from heterotrimeric αβγ G-protein complexes following binding of agonist ligands to GPCRs (e.g., epinephrine in the case of β adrenoceptors) and binds to and activates AC. The βγ subunits can also stimulate some AC isoforms. cAMP generated as a consequence of AC activation can activate several effectors, the most well studied of which is cAMP-dependent protein kinase (PKA)
When stimulated, a G protein dissembles into an alpha subunit and beta-gamma complex, both of which can directly regulate the activity of target protein in the plasma membrane.
Some GPLR activate adenylyl cyclase, thereby altering the intracellular concentration of intracellular mediator cAMP. A rise in cAMP level affect cell mainly by stimulating protein kinase A (PKA). PKA phosphorylates specific target protein and thereby alter the activity of protein. In this way these target protein enable the cell to make response to the small intracellular mediator.
