How to Diagnose Faulty Coil Pack without using a computer or OBD2 scan tools Check Engine Light. No special tools needed, just basic hand tools.
A misfire from one or more cylinders can be caused by many reasons from a faulty ignition system, fuel system or internal engine failure. Many times P0303 occurs when there are worn out spark plugs, spark plug wires, or a faulty ignition coil.

The simplest way to find a misfire on any Toyota running coil on plug ignition system, sometimes you can't just disconnect the plug lead as the ECU will shut the engine off if it detects a connection fault. this may not apply to all cars but still is one of the easiest ways to check a dead cylinder. i had been running this car for about 5 mins or so, and you can see the unburnt fuel building up in the Pre Cat converter and detonating in it, you can just see it through the join in the heat shields.
