Rolling circle mode of DNA replication is a commonly used method to replicate viral DNA molecules, which are circular and often single stranded. This also applies to episomal DNA in bacteria like plasmids, since a lot of them tend to be viral derivatives. Rolling circle replication is also relevant to the method of RCA - rolling circle amplification. In this video, I discuss the contrast between theta and rolling circle mode of DNA replication. The majority of the video focuses on detailing the mechanism by which rolling circle occurs and we discuss two specific examples - M13 and PhiX174 bacteriophages - to conceptually understand the mechanism, and how it can vary between different DNA molecules.
