Automated DNA Sequencing | Ibad Biology

Automated DNA sequencing is based on basic sequencing method developed by Fredrick Sanger.
Modified nucleotides called dideoxy nucleotides are used as chain terminators during DNA synthesis.
In automated sequencing the chain terminators or dideoxy nucleotides (ddNTPs) are tagged with different fluorescent molecules.
Template DNA i.e. DNA to be sequenced is obtained in single stranded form and mixed with normal DNA nucleotides (dNTPs), primers, DNA polymerase and one chain terminator.
In this manner DNA synthesis is carried out in four separate reaction tubes each containing a different chain terminator (dideoxy nucleotide).

All the reaction products are mixed and then separated through capillary gel electrophoresis according to their sizes.

A laser source is used to illuminate the fluorescent molecules as they pass by and the color is detected by detector.
The color information is converted to DNA sequence by computer.
