Types of Hemolysis | Alpha Beta and Gamma Hemolysis Detection | Basic Science Series
1. How to inoculate bacteria on blood agar plate:
• Grow bacteria overnight (use a fresh inoculum)
• Inoculate blood agar plate using your favorite streaking method
• “After overnight growth 37 C
• Held up to a light source light coming from behind
2. Beta Hemolysis
• Complete or true lysis (A clear zone)
• Toxic by-products
• Destroy red blood cells
• Multiple toxins !!!
• Beta hemolytic Streptococcus species, Streptococcus pyogenes
• Hemolysin that is oxygen labile Streptolysin O (low oxygen level)
• Anaerobic environment by inserting the inoculating loop vertically in the media
• (Stabbing)
• Oxygen stable hemolysin Streptolysin S (normal oxygen level)
3. Alpha Hemolysis
• Red blood cell hemoglobin
• Methemoglobin
• Fe³⁺ state, not the Fe²⁺
• Green or brown discoloration (around the colony)
• Cell membrane is intact (partial hemolysis)
4. Gamma Hemolysis
• Self-contradictory
• Non-Hemolytic = No reaction in the surrounding medium
• Non-hemolytic after 24 hours (weak alpha hemolysis)
• Enterococcus faecalis “Gamma streptococcus”
