In digital PCR assays, partitioning statistics are used to determine sample concentrations and absolute copies of the target molecule. The average number of targets per partition depends on the sample concentration and the number of partitions. The Poisson distribution governs the probability that a partition will contain a certain number of copies of the target. In this video, we walk you through the mathematical computation of the Poisson law for a digital PCR experiment using tennis balls and buckets.
The video also explains the rule of three and LOD.
