Note: be able to distinguish between eukaryotic and prokaryotic versions of some of these enzymes. DNA replication is unique for the MCAT (and biology in general) in that the prokaryotic enzymes (DNA Pol III, for example) are the main teaching points rather than their eukaryotic counterparts.

TL;DR: DNA replication involves a variety of associated proteins and enzymes that you should be familiar with!

Today’s MedCat video covers DNA replication, specifically discussing the role of origins of replication, replication forks, helicase, single-stranded binding proteins (SSBs), DNA topoisomerases (DNA gyrase in prokaryotes), primases, DNA Polymerase III, DNA Polymerase I, and finally DNA ligase. Further, we take a look at the DNA itself, assigning each of the four strands as either a leading strand with continuous DNA replication in the 5’ to 3’ direction or a lagging strand with multiple Okazaki fragments.

We wrap up with a AAMC-derived practice problem that tests your knowledge of DNA replication.
