Topo cloning also called..Topoisomerase based
Topo cloning also called..Topoisomerase based cloning is a DNA cloning method that does not use restriction enzymes or ligase, dosnt require post-PCR procedures.
The technique relies on the basic ability of complementary bases adenine (A) and thymine (T) to hybridize.
This post focuses on "sticky end" TOPO cloning;
however, the TOPO cloning technique has also bern adapted for blunt end cloning.
The key to TOPO cloning is the enzyme DNA topoisomerase I, which functions both as a restriction endonuclease and as a ligase by cleaving and rejoining supercoiled DNA ends to facilitate replication.
