Correlative AFM and STED – Rupture of microtubules in real-time

The video demonstrates a rupture of selected microtubules within a living human skin fibroblast cell. The microtubules are manipulated with the JPK NanoWizard® Atomic Force Microscope (AFM) and imaged with Stimulated Emission Depletion (STED) microscopy. The microtubules were labelled with SiR Tubulin and imaged with an Abberior Instruments Expert Line STED microscope (excitation, 640 nm; STED, 775 nm).
